Adding γ-CD to the sheath liquid enables the utilization of its hydrophobic cavity form inclusion complexation-based capture of the SDS molecules by their hydrophobic tail. Since the resulting cyclodextrin−SDS inclusion complexes were not observed in the MS spectra, this approach readily enabled the long-sought SDS-CGE of proteins with MS detection.

SDS CAPILLARY AGAROSE GEL ELECTROPHORESIS (SDS-CAGE) ANALYSIS OF LOW MOLECULAR WEIGHT PROTEIN STANDARDS, COUPLED WITH ESI-MS DETECTION. THE RESULTING MS SPECTRA ARE CLEARLY INTERPRETABLE FOR EACH PROTEIN. THE LEFT PANEL SHOWS THE TOTAL AND EXTRACTED ION ELECTROPHEROGRAM, WHILE THE RIGHT PANEL THE MS SPECTRA.
SDS capillary agarose gel electrophoresis (SDS-CAGE) analysis of Insulin and Lysozyme coupled to ESI MS detection. Panel A depicts the Total Ion Electropherogram (TIE, upper trace) and the Extracted Ion Electropherograms (EIEs, lower trace) of the protein analytes. Panel B presents the resulting MS spectra.
